چکیده

A phosphatase specific for P-glycolate5 was purified from extracts of tobacco leaves until no other phosphatase activity could be demonstrated in the preparation (10). Other plants have now been examined for the presence of this phosphatase. A new isolation procedure for the enzyme was developed because the (NH4) 2SO4 fractionation procedure, which was used with tobacco leaf extracts, inactivated the enzyme when applied with most other tissue. The enzyme has been found in green leaves but not in roots or etiolated leaves. P-glycolate phosphatase is of interest because Pglycolate and glycolate are labeled rapidly during photosynthesis with C14 0, (1,2,3,11). Also glycolate rapidly appears outside of isolated chloroplasts during photosynthesis (3), and thus the phosphatase may be a part of a chloroplast permease system. If this were possible then the phosphatase should be either a part of the chloroplast or closely associated with the chloroplast in vivo. Data will be presented with isolated chloroplast which support these suggestions.

Phosphoglycolate phosphatase and 2,3-diphosphoglycerate in red cells of normal and anemic subjects.

Red cell phosphoglycolate phosphatase (PGP) and 2,3-diphosphoglycerate (2,3-DPG) were investigated in normal and anemic patients and rabbits. In hemolytic anemia and blood-loss anemia, characterized by a young red cell population, there was an increase in both phosphoglycolate phosphatase activity and 2,3-diphosphoglycerate levels. In aplastic anemia, the phosphoglycolate phosphatase activity w...

3-Phosphoglycerate Phosphatase in Plants: II. Distribution, Physiological Considerations, and Comparison with P-Glycolate Phosphatase.

3-Phosphoglycerate phosphatase and phosphoglycolate phosphatase were found in leaves of all 52 plants examined. Activities of both phosphatases varied widely between 1 to 20 micromoles per minute per milligram chlorophyll. Plants were grouped into two categories based upon the relative ratio of activity of 3-phosphoglycerate phosphatase to phosphoglycolate phosphatase. This ratio varied between...

An essential developmental function for murine phosphoglycolate phosphatase in safeguarding cell proliferation

Mammalian phosphoglycolate phosphatase (PGP) is thought to target phosphoglycolate, a 2-deoxyribose fragment derived from the repair of oxidative DNA lesions. However, the physiological role of this activity and the biological function of the DNA damage product phosphoglycolate is unknown. We now show that knockin replacement of murine Pgp with its phosphatase-inactive PgpD34N mutant is embryon...

Structure- and function-based characterization of a new phosphoglycolate phosphatase from Thermoplasma acidophilum.

The protein TA0175 has a large number of sequence homologues, most of which are annotated as unknown and a few as belonging to the haloacid dehalogenase superfamily, but has no known biological function. Using a combination of amino acid sequence analysis, three-dimensional crystal structure information, and kinetic analysis, we have characterized TA0175 as phosphoglycolate phosphatase from The...

A Photorespiratory Mutant of Chlamydomonas reinhardtii.

A mutant strain of Chlamydomonas reinhardtii, designated 18-7F, has been isolated and characterized. 18-7F requires a high CO(2) concentration for photoautrophic growth in spite of the apparent induction of a functional CO(2) concentrating mechanism in air-adapted cells. In 2% O(2) the photosynthetic characteristics of 18-7F and wild type are similar. In 21% O(2), photosynthetic O(2) evolution ...
